Title: Parameters of semen quality affecting sperm freezability in Pradu-hangdum Chicken

Abstract: Pradu-hangdum chicken is well known as Thai native chicken that has a special character taste of meat and it can adapt to Thailand's weather and resistance to harmful disease. The aim of the present study is to investigate the parameter that has an effect on the quality of semen storage in cold and freezing conditions. Semen from thirty mature Pradu-hangdum roosters was collected. The semen was then preserved in Blumberger Hahnen Sperma Verdunner (BHSV) extender, and investigated for longevity of sperm stored in cold conditions and the frozen in BHSV extender. Moreover, the high-quality pooled semen collected from 30 mature Pradu-hangdum roosters mixed with BHSV extender and 10 % cthylene glycol, as a cryoprotectant, was divided into 4 groups with 12 replicates to investigate the effect of the different freezing processes and containers, including 1) semen preserved in 0.25 mL straw, 2) semen preserved in 0.5 mL straw, 3) semen preserved in 2 mL cryovial tube, and 4) pellet preparation semen preserved in 2 mL cryovial tube. The present study used a f freezing process. The results of this study showed that the sperm of Pradu-hangdum chickens has a slender head, semen color was creamy white, semen volume was 0.22 +- 0.03 mL, sperm concentration was 5.24x109+ 0.58 sperm/mL. The mean of total sperm motility was highest when stored in BHSV extender at 1 h (85.20 % +- 2.58), and followed by 24 h (56.00%+- 3.51) and 48 h (36.33 % +- 3.60), respectively. The longevity of stored sperm was decreased significantly at 24 h and 48 h (P<0.05). The freezability of Pradu-hangdum semen also showed a significant decline from 81.45% + 1.9 to 57.02% + 1.03 sperm motility (P<0.05) but still within the acceptable range of sperm motility and kinetic parameters for insemination. In addition, the mean of all motion parameters includes total motility, progressive motility, progressive fast motility, progressive slow motility, and some of the kinetic parameters include DCL(Distance Curve Line), DSL(Distance Straight Line), ALH(Amplitude of Lateral Head Movement), BCF(Beat Cross Frequency), and HAC(Head Activity). All parameter of the frozen-thawed sperm that was preserved in cryovial tube had significantly higher than any other groups (P<0.05). In conclusion, the time after semen collection and the semen freezing containers were important factors that impact the quality of Pradu-hangdum chicken fresh semen storage in cold condition and cryopreservation respectively. The inspected of kinetic parameter before semen cryopreservation may be beneficial to cryopreservation in Pradu-hangdum chickens.
