Abstract: In the present, the bull production industries demand specific sexual bulls due to different business productivity requirements. Therefore, sexual separation of sperm is very important to the industry and livestock. One of the most popular methods of sexual separation is monoclonal antibody enhancement. This method does not affect to the quality of sperm that is desired sex and also has specificity with high sexual separation accuracy. This research aimed to design and fabricate microfluidic system for sexual separation of bull sperm. Microfluidic system is one of scientific devices that was minimized to increase the efficiency of installation in conjunction with other devices and is convenient to carry, but still accuracy and speed in analyzing the results. The microchannel is designed by Layout Editor program and created by Soft Lithography technique. That is used to cooperate with microelectrode, it performs as aid of sexual separation and conductor to release the electric field into microchannel. In our experiment, separating particles included magnetic beads, magnetic beads on monoclonal antibodies and sperm with the function of magnetic beads on monoclonal antibodies by controlling microfluidic system fluid flow rate at 2 μl/min and releasing of 2.5 volts electric field. It was found that this microfluidic device can separate magnetic beads and magnetic beads on monoclonal antibodies of 100% and 95.42%, respectively. For sperm separation test, the results were shown that sperm Y with positive ions added moved to negative electrical fluid channel as our expectation which 53.61% success rate. By comparing with natural pregnancy, our proposed method is approximately 3.61% better.
