Title: Production and characterization of nuclease-resistant RNA Aptamers specific to Streptococcus suis

Abstract: Streptococcus suis (S. suis) is a gram-positive capsular bacteria known as a zoonotic
pathogen in humans that can cause meningitis, endocarditis, and septic shock. Most cases
of S. suis infection in humans, especially serotype 2, were found in the northern region of
Thailand. The conventional method for S. suis identification is culture and biochemical
tests, which are time-consuming and sometimes show the controversy results leading to
the mis- or delay diagnostic. Nowadays, the rapid with the highly accuracy methods are
required. Recently, there are studies of the molecule named “aptamer”, a short
oligonucleotide (single-stranded DNA or RNA) that can specifically bind to the target
cell through their conformational structure. The conventional method for producing
aptamer is Systematic Evolution of Ligands by EXponential enrichment (SELEX). In this
study, the monoclonal nuclease resistant RNA aptamer, R8-su12, against S. suis SS 2,
strain P1/7 was established using the cell-SELEX technique. In the cell-SELEX process,
firstly, the nuclease-resistant RNA library; containing 40-nt random region to yield a
diversity of approximately 1014 molecules was produced. Then, it was mixed with 107
CFU/mL of target cells. The increasing of stringency in each round of the cell-SELEX
resulted in high affinity and specificity of achieved aptamers. The round eighth (R8) RNA
aptamer pool was selected for specificity testing. No cross-reaction was observed when
tested with S. pneumoniae ATCC 49619 and S. pyogenes ATCC 19615 (95% CI,
p < 0.05). After cloning and sequencing, four groups of RNA aptamer were classified
according to the homology of nucleotide sequences. Group 1 showed the highest
frequency in the number of homologous sequences (7/26). The monoclonal R8-su12 RNA aptamer was then selected for further study. This monoclonal RNA aptamer was 84-nt in
length and the secondary structure showed an external 4-base loop with one closing
helices and stems-loops structure. No cross-reaction was observed when tested with
S. pneumoniae ATCC 49619, S. aureus ATCC 25923, E. coli ATCC 25922, and
P. aeruginosa ATCC 27853 (95% CI, p < 0.05). Moreover, the monoclonal R8-su12
RNA aptamer can also bind to S. suis SS 1/2, 1, 9, and 14. Importantly, this aptamer clone
revealed the ability to inhibit the biofilm formation of S. suis SS 2, strain P1/7. The
monoclonal R8-su12 RNA aptamer specific to S. suis SS 2, strain P1/7 is one of the
candidate RNA aptamers that can be used in various diagnostic tools such as biosensors
or the ELONA assay to detect S. suis in clinical isolates. Additionally, our selected RNA
aptamer can be developed for use in research field as a ligand to find a new molecule
involved in S. suis biofilm formation, and may also help to investigate the mechanism of
biofilm formation. Furthermore, this monoclonal R8-su12 RNA aptamer can be
developed as candidate ligands for drug delivery. Eventually, the RNA aptamer specific
to S. suis constructed in this study was the first step of the research that could be applied
in several scenarios involving in the mechanism of infection, diagnostic, and treatment of
S. suis infection. At last, it could lead to the control, an early and efficient treatment of

Abstract: This study aimed to explore the potential of main compounds from Curcuma comosa Roxb. in leukemia treatment. The Diarylheptanoid 7-(3,4-dihydroxyphenyl)-5-hydroxy-1-phenyl-(1E)-1-heptene (compound 1 or DDHPH) was obtained after purification from ethyl acetate (F-EtOAc) fractional extract, while diarylheptanoid trans-1,7-diphenyl-5-hydroxy-1-heptene (compound 2 or DHH) was obtained after purification from hexane (F-Hex) fractional extract. Both compounds 1 and 2 exhibited strong cytotoxicity against various cancer cell lines, while remaining nontoxicity toward normal white blood cells. Additionally, compound 1 demonstrated notable antioxidant and anti-inflammatory properties.
This study focused on proteins associated with cell proliferation and cell death. WT1 protein, a marker for leukemic cell proliferation, was significantly decreased after F-EtOAC and compound 1 treatments in a time- and dose-dependent manner. Moreover, compound 1 was found to induce NPM localization, resulting in the upregulation of p53 leading to cell cycle arrest and apoptosis. On the other hand, compound 2 demonstrated a drug modulator property, enhancing the efficacy of chemotherapeutic agents in drug-resistant cells.
In conclusion, this study successfully purified the main active compounds from C. comosa Roxb., including compound 1 and compound 2. Abstract: Star fruit (Averrhoa carambola L.) is a seasonal fruit found in Thailand that contains antioxidative compounds, such as phenolics, saponins, flavonoid C-glycosides, tannin, and L-ascorbic acid (Vitamin C), especially sour type. In Thailand, there is also a sweet-type of star fruit (SF) that is popularly grown and sold in the market, yet it remains uncharted in terms of reported or researched in terms of comparing the differences between the two types of SF. Therefore, this study has the objective (1) to study the active compounds and anti-oxidant activity in sweet- and sour-type SF extracts in vitro and (2) to evaluate the antioxidant activity and physical capacity of elderly individuals through the combined supplementation of a sweet-type SF and honey product, along with a home-walking exercise.
The study is focused on evaluating the amount of various active compounds including total phenolics, total flavonoids, and Vitamin C (Vit C), anti-free radicals, and anti-oxidant activities between two types of SF. Subsequently, the sweet SF was developed into an industrial pilot model of SF juice mixed with honey, in order to investigate its effectiveness among elderly participants, with an average age range of 60 to 70 years. The study is divided into four groups: the supplement and walking exercise group (S+WG) (n = 11) was compared with the supplement group (SG) (n = 11), the walking exercise group (WG) (n = 12), and the control group (CG) (n = 12). The dosage of the supplement was designed at 20 g in 150 mL of warm water which was administered twice daily for 4 weeks. Whereas exercise was a prescribed moderate-intensity walking exercise for 30 minutes, 3 days per week. Before and after the 4-week trial, blood oxidative stress status such as glutathione (GSH), Vit C, total antioxidant capacity (TAC), malondialdehyde (MDA), and 6-minute walking distance (6MWD) were assessed.
The findings indicated that the sour-type SF extract exhibited elevated levels of total phenolics, total flavonoids, and Vit C compared to the sweet-type, as well as scavenging activity on 2,2-diphenyl-1-picrylhydrazyl (DPPH) and nitric oxide (NO). However, sweet-type SF showed higher activity in scavenging the 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS˚+). Moreover, the sweet-type SF displayed superior protective effects against the oxidation of GSH, protein carbonyl, and lipid peroxide formation. Conversely, higher doses of sour-type SF extract triggered pro-oxidant activity by escalating GSH oxidation, protein carbonyl, and lipid peroxide formation in vitro with dose-response. For the study involved elderly participants in four groups, notable differences became evident following the 4-week intervention. In the WG, GSH and Vit C levels decreased, while TAC, MDA, and 6MWD significantly increased when compared to baseline measurements. The SG resulted in elevated plasma Vit C and TAC levels, as well as a reduction in MDA levels, with no significant changes in GSH and 6MWD. Lastly, the S+WG led to a decrease in both GSH and Vit C levels, a slight reduction in MDA levels, and a significant increase in TAC and 6MWD. Conversely, the CG showed no statistically significant changes in any of the measured parameters.
In conclusion, this study demonstrates that the supplementation of products containing sweet-type SF mixed with honey effectively enhances antioxidant activity, Vit C, and TAC levels, while concurrently reducing lipid oxidation. Furthermore, the combination of this product into a regular home-walking exercise regimen proves to be beneficial in improving the walking capacity of elderly individuals.2023-09-06T00:00:00Z
